PARP, Laddering and other kits
The HT PARP/Apoptosis Assay is ideal for measuring the activity of PARP in cell extracts before and during apoptosis. This ELISA semi-quantitatively detects PAR deposited onto immobilized histone proteins in a 96-well format. An anti-PAR monoclonal antibody, goat anti-mouse IgG-HRP conjugate, and HRP substrate are used to generate a signal. Thus, signals correlate with PARP activity. Etoposide is a topoisomerase II inhibitor that stabilizes this enzyme after it cleaves DNA. It is included as a control apoptosis inducer.
With the TACS® Apoptotic DNA Laddering Kit DNA fragments are analyzed by horizontal gel electrophoresis followed by ethidium bromide staining. When analyzing tissue samples, it is necessary to disrupt the tissue and create a cell suspension. The Tissue Supplement Kit (Cat# 4859-20-K), available separately, provides the reagents to allow DNA laddering analysis in tissues.
FLISP, Fluorescent-Labeled Inhibitors of Serine Proteases assay kits detect changes in intracellular chymotrypsin-like enzyme activity in whole living cells. FLISP probes will interact with the active catalytic sites of chymotrypsin-like proteases, quickly forming covalent bonds with either the reactive site histidine (N-H) (with the CMK-type FLISP probes) or the reactive serine OH group (when DAP-containing FLISP probes are utilized).
